Vβ8T細胞による自己免疫病増悪機構の解明とTCRを標的とした遺伝子治療

研究概要	IL-12p40分子によるIL-12機能抑制効果を介する自己免疫症状の軽減を期待して、独自に開発した自己免疫病モデルマウスMRL-lpr^<cg>(lpr^<cg>)に、発生工学技術によりIL-12p40cDNAを導入して、IL-12p40-lpr^<cg>トランスジェニックマウス(Tgp40-lpr^<cg>)を作出した。Tgp40-lpr^<cg>ではp40が肝臓で強く、腎臓で弱く発現し、血中では非Tgマウスに比べて数千倍にも達していた。血中IgGレベルが顕著に抑えられ、代表的自己抗体である抗DNA抗体産生はIgM、IgGクラス及びIgG2aサブクラスで有意に抑えられたが、IgG 1サブクラスではむしろ上昇した。さらにINF-γ産生が抑えられなくなる傾向があり、Th1細胞活性の抑制が認められた。しかし、免疫複合体量にはほとんど影響がなく、臨床症状ではタンパク尿症のわずかな軽減と軽度の延命効果が見られたが、異常T細胞によるリンパ節腫脹や糸球体腎炎と血管炎に対しては全く影響を与えなかった。これは、大量のp40存在は確かにIL-12の機能を抑制する反面、IL-12産生を亢進するためと推察された。さらに、IL-1α/βを欠損させたIL-1ターゲットlpr^<cg>マウスを作出して、IL-1の役割を追求したところ、リンパ節腫脹は予期に反して顕著に促進され、糸球体腎炎、蛋白尿症などの臨床症状は全く改善されなかった。前年度の成果として、Vβ8^+T及びCD4^+T細胞の欠損が明瞭に臨床症状を軽減することを確認しているので、サイトカインよりもT細胞を標的とする遺伝子治療が効果的と考えて研究を進めているが、現在までに画期的な治療効果が得られていない。
