研究概要	マウスのmi遺伝子座にコードされている転写因子MITFの機能について調べた。mi遺伝子座の突然変異のうち最も著明な症状を示すのはmi/miマウスである。mi突然変異遺伝子によりコードされている蛋白mi-MITFはbasic領域に4個ならぶアルギニンのうちの1個が欠失するため、DNA結合能を完全に失うと共に、核移行能にも明らかな欠陥を有する。一方tg/tgマウスではMITF自身のコーディング領域の上流に移入された遺伝子が挿入されたために、MITFがまったく発現されなくなる。tg/tgマウスの症状はmi/miマウスに似るが、その程度はmi/miマウスの方が重症である。mi/miとtg/tgの差を調べるため、+/+マウス、mi/miマウスtg/tgマウスの骨髄細胞を培養して、培養マスト細胞を得、おのおのの培養マスト細胞からcDNAライブラリーを作製した。(+/+-mi/mi)と(+/+-tg/tg)の2種のサブトラクションライブラリーを作ると、(+/+mi/mi)サブトラクション中には(+/+-tg/tg)サブトラクションライブラリーに比し多くのクローンが検出された。mi-MITFが発現する方が、MITFを全く発現しない場合に比し、発現する遺伝子が少ないわけで、mi-MITFは負の転写活性化能を持っていることがわかった。次にMITFがその転写に関係していることが知られている遺伝子について、ノーザンプロットによりmi/miマウスとtg/tgマウスでメッセンジャー(m)RNA量に差があるかどうか調べた。調べた遺伝子の大部分で、mi/mi培養マスト細胞におけるmRNA量はtg/tg培養マスト細胞におけるmRNA量より低く、サブトラクションライブラリーで得た結果と一致した。
