遺伝子操作マウスを用いた住血吸虫症ワクチン開発に関する基礎的研究

研究概要	住血吸虫症ワクチンの開発に資するため、遺伝子操作マウスを用いて住血吸虫感染に対する防御免疫機構について検討した。すでに、マウスにおいては、マンソン住血吸虫の感染に対し、γ線照射セルカリアの接種により不完全ながら防御免疫を誘導できることが知られており、IFNγやNOの関与の可能性が報告されてきている。そこで、Th1応答に障害のみられることが予想されるIRF-1ノックアウト(KO)マウスを用いて、マンソン住血吸虫感染に対する防御免疫について検討した。その結果、KOマウスでは有意な感染防御が観察されないことが示された。脾細胞の免疫応答では、野生型(WT)と比較した場合、Th2(IL-4、IL-10)の2次応答は同程度に起こるが、IFNγの2次応答は顕著に低下していることが判明した。また、産生されたサイトカイン総体としてのマクロファージ刺激能を、シストソミュラ傷害性およびNO産生で検討したところ、いずれも顕著に低下していた。さらに、組換えIFNγでの刺激実験により、マクロファージ自体のシストソミュラ傷害能も低下していることが明らかになった。液性免疫では、IgG抗体産生が低下していた。以上の事実から、転写因子IRF-1はマンソン住血吸虫感染に対する防御免疫において必須であり、IRF-1KOマウスではさまざまなレベルで虫体に対する免疫応答が低下した結果、感染防御の消失が観察されることが明らかになった。
