研究概要	サイトカインシグナルを直接核に伝達するJak-STAT伝達系は、Th1,Th2分化やB細胞の抗原応答を決定する基本的なシグナル伝達系の一つとして重要であるだけでなく、発生初期の形態形成、器官形成、細胞の運動などさまざまな場で重要な作用をもっている。これまでIL-6による増殖、分化のいずれにもSTAT3転写因子が重要な役割をもつことを示してきた。今回、IL-6/gp130シグナル下流で、STAT3活性化に関わるセリン/スレオニンキナーゼ伝達路の全容を明らかにしていくことを目指して、解析を行い、以下の諸点を明らかにした。(1)IL-6刺激により活性化されるSTAT3キナーゼには、高濃度IL-6でのみ活性化されるMEK阻害剤感受性キナーゼ系と低濃度IL-6でも活性化される阻害剤H7に感受性のキナーゼ系があることを示し、後者のSTAT3キナーゼ系が、Ras/MAPK活性化の観察されない細胞や生理的濃度での刺激の際にも活性化されることから、実際に重要な役割を担うことを示唆した。(2)gp130受容体細胞内領域のうちSTAT3ドッキング領域として重要なpYXXQモチーフそのものが、H7感受性STAT3キナーゼ活性化に必要十分なモチーフであることを見い出した。(3)このpYXXQ由来伝達系を介したH7感受性STAT3キナーゼ伝達系は、Erk,p38,PKCδなどのキナーゼ系とは異なるものであること、さらにH7感受性伝達路によるSTAT3のセリンリン酸化は核内でも認められることを示した。この新規伝達系の構成要素の同定、役割の解明は今後の課題である。(4)継続課題として、stat3遺伝子プロモーター上に存在する低親和性STAT3結合配列に変異を導入したプロモーターノックインマウスを作製し、STAT3によるstat3遺伝子活性化が組織特異性をもつことを示した。この自己遺伝子活性化ループの役割は検討中である。
