高発癌感受性集団や一般的発癌に対する特異的予防法の開発

研究概要

本年度は、別紙の研究業績にあるように、発癌抑制や癌患者の予後に関係する重要な発癌抑制関連遺伝子であるcdk inhibitorであるp27遺伝子プロモーターを発癌抑制効果の知られるVitamin D_3(VD_3)が活性化することにより、細胞増殖抑制効果を示すことを明らかにした。興味深いことに、VD_3はVD_3受容体を介さずに、Sp1とNF-Yを介してp27プロモーター活性を活性化することを見いだした。この結果は、VD_3の副作用として重要な高カルシウム血症の要因となるVD_3受容体を介さない、すなわち高カルシウム血症のない薬剤開発の可能性を示すデータとして興味深い。また、VD_3が大腸癌などに対する予防効果が知られているが、そのメカニズムとして今回の経路が重要である可能性が高い。さらにリ・フラウメニ症候群のようにp53遺伝子が遺伝的に失活していて発癌リスクの高い人に対して、その標的遺伝子であるWAF1をプロモーターレベルで活性化することが予防に有用である可能性を示し、酪酸などのヒストン脱酵素阻害剤が有用であることを報告してきたが、さらに今回それにWAF1プロモーターのSp3が重要であることを見いだした。
