自殺遺伝子とサイトカイン遺伝子の同時発現による消化器癌のin vivo遺伝子治療

研究概要

1) 胎児性癌抗原(CEA)産生ヒト大腸癌に選択的な遺伝子発現を誘導するために、CEA高産生ヒト大腸癌より、CEAプロモーター領域をPCR法を用いて増幅し特異性を検討したところ、204 bpおよび419 bpより成るCEAプロモーター領域は共に、CEA産生ヒト大腸癌選択的な外来遺伝子発現を誘導し得た(Int J Cancer 1998;78:242-247)。
2) CEA産生大腸癌細胞をヌードマウスの腹腔内に接種し大腸癌の腹膜播種モデルを作製し、CEAプロモーター+CD遺伝子を有するレトロウイルスの産生細胞を腹腔内に接種した後に5-FCを投与すると、マウスの生存日数は有意に延長した。さらに、SV40プロモーター+CD遺伝子を有するレトロウイルスの産生細胞を接種したマウスでは、5-FC投与により著明な骨髄抑制を来したが、CEAプロモーター+CD遺伝子を有するレトロウイルスの産生細胞を接種した群では、骨髄抑制を認めなかった(Gene Ther 1999;6:83-90)。
3) レトロウイルスのパッケージング細胞にポリオーマウイルスの初期遺伝子領域を導入すると、レトロウイルスの産生効率は約100倍上昇し、高力価なレトロウイルスの産生細胞を作製し得た(HumGene Ther 1998;9:161-172)。
4) レトロウイルスを用いて単純ヘルペスウイルスのチミジンキナーゼ遺伝子を導入した肝癌細胞をマウスに移植すると、導入遺伝子のmethylationがおこりガンシクロビル(GCV)に耐性を示したが、5-azacytidine処置により遺伝子発現は回復し、GCVに感受性を示した(Gene Ther 1998;5:1299-1305)。
5) アデノウイルスは、in vivoにおいても効率の高い遺伝子導入が可能なベクターであるが、抗アデノウイルス抗体陽性のヒト血清と混合した後にラット門脈より投与すると、遺伝子導入は誘導されず、抗アデノウイルス抗体陽性のヒトでは、アデノウイルスの血管内投与による遺伝子導入は難しいと考えられた(Anticancer Res 1998:18:2345-2352)。
