自殺遺伝子とサイトカイン遺伝子の同時発現による消化器癌のin vivo遺伝子治療

研究概要

1)胎児性癌抗原(CEA)プロモーターの制御下に、大腸菌由来のシトシンデアミナーゼ(CD)遺伝子を発現するレトロウイルスをマウスに腹腔内の注入しても、CD遺伝子の骨髄での発現は認められず、5-FCを投与しても副作用を認めなかった。一方、SV40プロモーター+CD遺伝子のレトロウイルスを腹腔内に注入したマウスでは、骨髄においてCD遺伝子発現が誘導され、5-FCを投与すると著明な骨髄抑制を来し死亡した(Cancer Gene Ther 6,572-580,1999;Int J Oncol 15,107-112,1999)。
2)単純ヘルペスウイルス由来チミジンキナーゼ(TK)とCD遺伝子の抗腫瘍効果には、T細胞を介した宿主の腫瘍免疫が重要な役割を演じており、正常なT細胞系免疫能を有さないヌードマウスでは、TKあるいはCD遺伝子を用いた癌に対する遺伝子治療効果は著しく抑制された。一方、正常な免疫能を有するマウスでは、皮下に形成された肝癌内にTK遺伝子を含むレトロウイルスを反復注入しガンシクロビルを投与すると、すべてのマウスにおいて皮下肝癌の完全消失を誘導し得た(Int J Cancer 81,592-597,1999;Int J Cancer 83,374-380,1999;Scand J Gastroenterol 34,1033-1041,1999;Gene Ther 6,525-533,1999)。
3)アデノウイルスは免疫反応を惹起するため遺伝子の再発現が困難であるが、アデノウイルスの再投与前に免疫抑制剤であるシクロホスファミドを投与しておくと、遺伝子の再導入および再発現の誘導が可能であった(Gene Ther 6,749-757,1999)。
4)癌の発育には血管内皮増殖因子(VEGF)の発現が不可欠であるが、VEGFの受容体であるKDR/Flk-1の中和抗体あるいはプロテインキナーゼC特異的阻害剤を投与すれば、腫瘍内の血管新生が著明に抑制され腫瘍発育も抑制された(Hepatology 30,1179-1186,1999;Cancer Res 59,4413-4418,1999)。
