自殺遺伝子とサイトカイン遺伝子の同時発現による消化器癌のin vivo遺伝子治療

研究概要

1)組換えアデノウイルスを経静脈的に注入すれば、正常肝マウスのみならず、劇症肝炎あるいは肝硬変モデルマウスにおいても、肝臓へ安全かつ効率的に外来遺伝子の発現を誘導し得ることを示した(Gut47,563-570,2000)。アデノウイルスの静脈内注入から1年後に、アデノウイルスを経静脈的に再注入しても、肝臓での遺伝子の再発現は誘導されないが、再注入時に免疫抑制剤であるFK506を投与しておけば、肝臓での遺伝子の再発現を誘導し得ることを示した(Int J Cancer 85,839-844,2000;Int J Oncol 18,575-580,2001)。アデノウイルスをマウスの皮下肝癌内に直接注入すれば、腫瘍内への効率の良い遺伝子導入が可能であった。また、アデノウイルスを腫瘍内に再注入した場合には、初回投与時の1/10程度の遺伝子発現を誘導し得ることを示した(Int J Oncol 18,57-65,2001)。
2)SV40やレトロウイノレスのlong terminal repeatなどのウイルス由来のプロモーター、さらにphosphoglycerate kinaseなどのhousekeeping遺伝子のプロモーターに、α-fetoprotein遺伝子のenhancerを含む上流のsequenceを接続したキメラ制御因子を用いれば、自殺遺伝子やサイトカイン遺伝子などの治療遺伝子を肝癌細胞に選択的に発現させ得ることを示した(Int J Cancer 87,247-252,2000;Eur J Cancer 37,140-147,2001)。
3)遺伝子銃を用いれば、マウス肝臓への広範な遺伝子導入が可能であり、導入遺伝子の発現は、漸減しながらも4週間持続することを示した(Gene Ther 7,1132-1136,2000)。
4)肝癌の癌性腹膜炎モデルマウスの腹腔内に、自殺遺伝子を組み込んだレトロウイルスを直接注入しプロドラッグを投与すれば、マウスの生存日数は有意に延長し、その際に少量のIL-2を併用投与すれば、さらに有効な抗腫瘍効果を得られることを示した(Scand J Gastroenterol 35,852-860,2000)。
