血管壁リモデリングに及ぼすトロンビン/トロンビン受容体の関与とそのシグナル伝達系

研究概要

本年度の研究で明らかにしえたことは以下の通りである。
1.トロンビンはトロンビン受容体の1型(PAR-1)を活性化して、その後MAP-キナーゼカスケードを経て、NF-κBの活性化し、結果として炎症性サイトカイン、細胞接着因子、組織因子、PAI-1などの発現を強めるが、本年度は新たに、核内シグナルの統合因子であるCBP/p300を活性化することを見出した。
2.CBP/p300の活性化の後、本因子がリジンサイトのアセチル化活性があるため、細胞核内にはリジンアセチル化蛋白が集積した(これをhypernuclear acetylation,HNA)と命名した。
3.HNAは冠動脈粥腫部位で血管平滑筋細胞にin vivoでも証明し得た。
4.リジンアセチル化の抑制で、トロンビンによる血管平滑筋細胞の増殖は抑制された。
5.これらのことからHNAは一部トロンビンで誘導され、これは動脈硬化の発症に深く関係しているものと考えられた。
今後の重要な課題として、トロンビン→PAR-1→MAPキナーゼカスケード→CBP/p300→細胞増殖のいずれかのステップでシグナルを遮断して、in vivoで動脈硬化を抑制する戦略の開発が重要であろう。
