臍帯血造血幹細胞制御の分子機構の解明とその無血清体外大量培養法への応用

研究概要

1. マウス胎生期aorta-gonad-mesonephros(AGM)領域由来ストローマ細胞株AGM-S3の樹立我々は、臍帯血造血幹細胞の無血清体外大量培養法を確立するためには、胎生期造血に由来する臍帯血造血幹細胞の増殖機構を明らかにすることが重要と考え、胎生期マウスのAGM領域に注目した。胎生期マウスの造血幹細胞は、胎生10日のAGM領域に発生し、胎生11日にかけて著明に増幅する。従って、この時期のAGM領域には胎生期造血幹細胞を増幅する分子が発現されていると考え、胎生10.5日のAGM領域よりストローマ細胞株AGM-S3を樹立した。
2. AGM-S3の造血支持能の検討
AGM-S3の造血支持能を検討するために、蛍光活性化細胞分離装置で分離されたLin^-c-Kit^+Sca-1^+細胞とLin^-c-Kit^+Sca-1^-細胞をAGM-S3細胞上で共培養したところ、培養3日目のコブルストーン形成及び培養10日目のコロニー形成細胞、脾コロニー形成細胞産生はLin^-c-Kit^+Sca-1^+細胞との共培養系においてのみ認められた。このことより、AGM-S3細胞のマウス造血支持能は主に未分化なLin^-c-Kit^+Sca-1^+細胞に作用すると考えられた。さらに、Lin^-c-Kit^+Sca-1^+細胞中の長期造血再構築能を有する造血幹細胞をマウス移植系を用いて検討したところ、AGM-S3上で10日間共培養されたLin^-c-Kit^+Sca-1^+細胞中にも造血幹細胞は維持されていた。以上の結果より、マウスAGM領域由来ストローマ細胞株AGM-S3は、造血幹細胞を含む造血細胞を支持する能力を有すると考えられた。
