造血幹細胞の発生における分子基盤の解明

研究概要	マウスでは造血幹細胞、血管内皮細胞の共通祖先細胞であるヘマンジオブラストは胎生6.5日目、側板中胚葉領域で発生し、傍大動脈臓側板中胚葉領域(以下P-Sp領域と略す)に移動し造血幹細胞、血管内皮細胞に分化する。本研究において胎生9.5日目にチロシンキナーゼ受容体であるc-KitおよびTIE2陽性の造血幹細胞がP-Sp領域の近傍の臍腸間膜動脈内で血管内皮細胞に接着、増殖することを明らかにした。TIE2陽性造血幹細胞は、そのリガンドであるアンジオポエチン-1刺激により、インテグリンを介した細胞接着性が亢進することを見い出した。さらに造血幹細胞の自己複製の系にTIE2-アンジオポエチン-1のオートクリンループが関与することが次第に明かとなってきている。上記の研究過程で造血幹細胞の発生メカニズムを解析するためにP-Sp領域をマウスより単離、培養することにより、造血幹細胞および血管内皮細胞の両者を発生、分化させることに成功した。本培養では造血幹細胞は臍腸間膜動脈内で観察されたように、血管内皮細胞に接着し増殖する。本培養を利用し血管内皮細胞は造血幹細胞の発生、増殖に必須のストロマ細胞として機能することを明かにした。またマウスTIE2の細胞質内領域の細胞膜貫通部直下に相当する848番アミノ酸のArgをコードするCGGをTGGとpoint mutationをいれ、アミノ酸をTrpに変換し、この変異TIE2 cDNAをマウスB細胞株Ba/F3細胞に導入したところ、TIE2のリガンドであるアンジオポエチン-1の刺激なしにTIE2受容体はダイマー形成が誘導され、さらにその自己燐酸化が観察された。この変異TIE2 cDNAをレトロウイルスベクターに組み換え、フローサイトメトリーにより回収したTIE2陽性造血幹細胞に感染させ造血幹細胞の未分化性を長期に維持できるか否かを今後検討する。
