研究概要

本研究はグルココルチコイド受容体機能の制御機構を分子レベルで明らかにし、グルココルチコイドによる遺伝情報発現調節機構を、研究成果の臨床応用をも視野にいれて包括的に解明することを目的とする。
[1] グルココルチコイド受容体の翻駅後修飾
・グルココルチコイド受容体分子中のレドックス感受性アミノ酸を同定しつつある。
・グルココルチコイド受容体と相互作用する細胞内還元酵素がチオレドキシンであることを同定した。
[2] グルココルチコイド受容体の核移行の分子機構解明
・グルココルチコイド受容体分子中の核移行シグナルと会合する細胞質内蛋白がimportin-αであることを同定した。
・グルココルチコイド受容体の核移行のレドックス制御機構を解明した。
・グルココルチコイド受容体の核から細胞質への輸送機構を究明中である。
[3] クロマチンの高次構造とグルココルチコイドによる転写調節機構
・グルココルチコイド受容体と相互作用するcoactivator,corepressorのの機能調節機構ならびにクロマチン高次構造に与える薬剤のグルココルチコイド依存性転写に対する影響を検討中である。
[4] NF-kBとの相互作用の解明
・NF-kBのcofactorのcDNAクローニングに成功し、その生化学的特性を解析中である。
