心血管系での誘導型NO合成酵素遺伝子の発現調節と作用の分子機構

研究概要	血管平滑筋細胞(VSMC)ではサイトカインによって誘導型NO合成酵素(iNOS)が発現するが、その情報伝達経路と過剰生成NOの機能は不明である。培養ラットVSMCを用いてサイトカイン(IL-1,TNF-α)およびスフィンゴミエリナーゼ(SMase)によるNF-kB活性化経路とiNOS発現との連関を分子レベルで解析した。IL-1やTNF-αはNF-kBを活性化してiNOS誘導をもたらし、この経路にはIkBキナーゼの活性化によるIkBBαのリン酸化とそれに引き続くIkBBαのプロテアソームによる分解を介して、NF-kBによるiNOS遺伝子の転写亢進によることを明らかにした。一方SMaseはNF-kBを活性化してiNOSを誘導するが、その効果はTNF-αと相乗的に作用する。SMaseはkBαのリン酸化や分解をもたらさないことから、IkBキナーゼとは別の経路を介したNF-IkB活性化をもたらすものと推定される。外因性NOドナー(NOR-3)は、サイトカインで発現したiNOSを転写レベルで抑制し、直接IkBαのリン酸化阻害をもたらすことを明らかにした。したがってiNOSで過剰生成されたNOはその発現を自ら抑制するというオートフィードバック機構が存在するものと推定される。更に、iNOS遺伝子をVSMCにトランスフェクトして過剰発現させると、著明な細胞の自己死(アポトーシス)をもたらすことをTUNEL法やDNAラダー形成によって証明した。したがってVSMCでのiNOS誘導はアポトーシスを介して血管リモデリングに関与することが示唆される。
