研究概要

本研究ではin vitroのリンパ球混合培養の系において副刺激を遮断することによって抑制性T細胞が誘導されるかどうか、この細胞がin vivoの移植の系においても拒絶反応を制御できるかどうかを検討した。まず、T細胞の活性化においてB7-CD28/CTLA-4とICAM-1/LFA-1を介する副刺激の経路が大きく関わっていることを、それぞれの分子に対する特異抗体をもちいたマウス膵島移植の系において明らかにした。次に、BALB/c(H-2^d)をstimulator、C57BL/6(H-2^b)をresponderとし、抗B7-1抗体、抗B7-2抗体の両抗体存在下に5日間培養することによってアロ抗原非特異的なアナジー細胞の誘導に成功した。この細胞はConA刺激に反応できたが、抗CD3抗体刺激に対する反応性は低下していた。また、アロ抗原に対する反応性はIL-2(100U/ml)の存在により回復することが明らかとなった。これらのアナジー細胞をprimary MLRの系に添加すると、照射の有無に関わらず、細胞数依存性に抗原非特異的にアロ抗原に対する反応を抑制した。In vivoの効果はBALB/cの膵島をストレプトゾトシン糖尿病C57BL/6に2.5Gy照射後移植する系において検討したところ、in vitroで作成したこのアナジー細胞4x10^7個を移植時静注すると、同数の無処置群、あるいはコントロール抗体処理脾細胞投与では急性拒絶が誘導されたのに対し、有意な生着延長がえられ、5例中2例は100日以上長期生着した。すなわち、in vitroで副刺激を遮断することによって得られたアナジー細胞がin vivoにおいて拒絶反応を抑制しうることを示す初めての知見が得られた。
