網膜芽細胞腫遺伝子キメラマウスを用いた骨肉腫発癌機構の解明

研究概要	網膜芽細胞腫(Rb)遺伝子を欠損するES細胞(MIT.Tylar Jacks博士より供与)を用いて、Rb(+/+)/(-/-)のキメラマウスを作成した。このマウスの胎仔あるいは新生仔より頭蓋骨及び長管骨を単離し、コラゲナーゼ及びディスパーゼを用いて細胞を分散、培養を開始した。単離した細胞は、通常の培養方法に加えて、培地にG418を添加して、Rb(-/-)のクローンを選択的に増殖するような条件でも培養した。得られたクローンの遺伝子型の確認は、アレル特異的なPCRを用いて行った。通常の培養方法のクローニングでは、10個のクローンが得られたが、そのうちRb(+/+)のクローンは1クローンのみで、他はRb(-/-)であった。G418添加の培養条件では、頭蓋冠及び長管骨よりそれぞれ29個のクローンが得られ、遺伝子型は全てRb(-/-)であった。この結果からは、Rb(-/-)の骨芽細胞は何らかのinvitroにおけるgrowth advantageを獲得している可能性が示唆される。頭蓋骨より得られた細胞のALP活性は、マウス線維芽細胞よりは高値であったが、代表的な骨芽細胞であるMC3T3E 1より低かった。一方長管骨より得られた細胞のALP活性は、ほとんど認められないものから、MC3T3E1の約10倍の値を示すものまであった。以上の全てのクローンにおいて、KT-PCRにより骨芽細胞系のマーカーであるCbfa1及びosteocalcinの発現が確認できた。Rb遺伝子の機能の一つとして、分化制御があげられているが、以上の結果は少なくとも骨芽細胞系の分化マーカーの発現に関しては、Rb欠損は影響を与えないと考えられる。現在、これらの細胞系にテトラサイクリンにて発現誘導可能なヒト変異型p53遺伝子を導入し、その解析を行っている。
