子宮に発生する良性・悪性平滑筋腫瘍の腫瘍性に関する包括的研究

研究概要	本年度は、1)正常子宮平滑筋、子宮筋腫、平滑筋肉腫の各組織においてestrogen receptor(ER)、progesterone receptor(PR)の発現、また腫瘍抑制遺伝子p53について蛋白質、RNAレベルにおいて定量的、定性的に検討したところ、正常子宮平滑筋および子宮筋腫細胞には、ER、PRは共に強く発現し、p53の発現はほとんど見られなかった。一方、平筋筋肉腫についてはER、PRの発現抑制がみられ、これに伴ってp53の過剰発現が見られ、p53geneのmissense mutationは4/14(28.5%)に観察され、平筋筋腫瘍の悪性化に性ステロイド受容体発現の異常とp53の異常が関与していることが判明した。ついで、2)子宮平滑筋の中に発生する良性腫瘍の子宮筋腫と悪性腫瘍の平滑筋肉腫は子宮平滑筋に比べると細胞の走行性や、形態に違いがあり、アクチン結合蛋白質であるカルポニンh1が平滑筋肉腫で発現低下していた。そこで、子宮平滑筋肉腫細胞株SKNにカルポニンh1遺伝子を導入したところ、細胞が正常平滑筋に類似した配列を示すようになり、増殖能・造腫瘍能ともに抑制された。カルポニンh1はヒト子宮平滑筋肉腫において腫瘍抑制因子としての役割を果たしている可能性が示唆された。また、子宮平滑筋肉腫細胞においては、細胞接着や細胞形態に関わるシグナル伝達がカルポニンh1により制御されている可能性と将来的なカルポニンh1遺伝子導入による平滑筋肉腫の治療応用を示唆した。
