女性生殖器細胞のアポトーシス調節機構:その局所・内分泌因子と細胞間相互作用の解析

研究概要

CDDPによるアポトーシス誘導経路を検討する目的で、CaSki細胞におけるCDDP添加後のFas/Fas ligand発現態度を検討した。CaSki細胞の無血清培養系にCDDPを添加し、CaSki細胞でのFas蛋白の発現に及ぼすCDDPの影響を免疫染色法ならびにWestern blot法で経時的に検討し、Fas ligand蛋白の発現に及ぼすCDDPの影響を免疫染色法で経時的に検討した。また、CDDP添加6時間後のCaSki細胞からtotal RNAを抽出し、CaSki細胞のFas ligand mRNA発現に及ぼすCDDPの影響をNorthern blot法で検討した。CaSki細胞の細胞質にはFas蛋白とFas ligand蛋白の局在を認め、CDDP添加24時間後と48時間後において、添加群では非添加群に比して、CaSki細胞内Fas蛋白とFas ligand蛋白の発現が共に増加することを認めた。またCaSki細胞内には2.0KbのFas ligand mRNAの発現を認め、CDDP添加群では非添加群に比しFas ligand mRNA発現が増加することを認めた。CDDPはCaSki細胞でのFas/Fas ligand蛋白ならびにFas ligand mRNAの発現を増加させたことより、CDDPによる子宮頚部扁平上皮癌細胞のアポトーシス誘導は、Fas/Fas ligand系を介して起こることが示唆された。
他方、多くの悪性腫瘍細胞ではテロメラーゼ活性の上昇によって細胞寿命が延長することが知られるが、抗癌剤の抗腫瘍効果とテロメラーゼ活性との関連については明らかでない。そこで子宮頚部扁平上皮癌細胞株(CaSki)のin vitro培養系で、高用量CDDP添加に伴う細胞増殖能、アポトーシス発現、ならびにテロメラーゼ活性に与える影響を明らかにしようとした。その結果、CDDP添加6時間後より培養CaSki細胞のBrdU陽性率は低下し、アポトーシス陽性細胞率が増加した。他方、細胞数はCDDP添加48時間後より減少を示した。CaSki細胞のテロメラーゼ活性はCDDP非添加群では不変であったのに対し、CDDP添加群では添加12時間後より低下の傾向を示し、CDDP添加48時間後には有意に低下することを認めた。CDDPは、添加6時間後よりCaSki細胞増殖能の低下と同時にアポトーシスを惹起させ、CDDP添加12時間後よりテロメラーゼ活性を低下させた。これらの事実より、CDDPの抗腫瘍効果は、添加後早期に起こるアポトーシス、並びにアポトーシスに遅れて起こるテロメラーゼ活性低下に伴う分裂寿命の延長抑制による可能性が示唆された。
