気道腺組織の分化と増殖についての研究

研究概要	本年度と過去4年間の研究成果の概要を示す。腺細胞の3D培養は無血清倍地での培養に成功し、これを用いて増殖因子(EGF,KGF)およびビタミンA誘導体の培養腺細胞の増殖と分化に及ぼす影響を明らかにした。培養腺細胞のムチン産生についても検討し、LPS刺激によりMUC2,MUC5ACの発現は増強されたがMUC5Bに変化をみなかった。ラット気管粘液産生モデルを用いて薬物の粘液産生量に及ぼす影響を影響を定量的測定し、ステロイド,14員環マクロライドの粘液産生抑制作用を明らかにした。また、鼻・副鼻腔患者における粘液産生に及ぼす影響を検討し、粘液産生が、急性,慢性炎症さらにI型アレルギー反応により産生される場合にいかなる相違が存在するかを検討した。また鼻・副鼻腔の病的状態で産生された粘液がどのような物理学的性質を有しており、その性質は何により決定されるかを明らかにした。結論として鼻・副鼻腔の慢性炎症では主として腺細胞からの粘液産生が亢進しており、この原因は増殖因子による腺の増殖と粘液産生を亢進する種々の炎症産物によるものと考えられる。腺の増殖に関与する因子は他にも存在する可能性があり、今後はさらにこれら因子の検討を行い、臨床において腺の過分泌抑制を可能にしたい。
