鼻副鼻腔粘膜の免疫アレルギー疾患にかかわるT細胞を標的とした治療の開発-分子生物学的手法を用いたヒト鼻粘膜T細胞の解析-

研究概要	1.アレルギー性鼻炎あるいは慢性副鼻腔炎患者鼻腔粘膜より採取した鼻粘膜リンパ球を用いて、フローサイトメトリーを行い、T細胞分画の解析を行った。その結果、種々の割合でCD4+ヘルパーT細胞ならびにCD8+抑制性T細胞が分布していることが示された。 2.PRISM7700sequence detectorを使用して、鼻腔粘膜より得られた少量のリンパ球を用いて、定量的なRT-PCRを行い、Th1タイプ(IFN-gamma,IL-2)およびTh2タイプ(IL-4,IL-5)のサイトカインについてmRNAでのサイトカインプロフィールについて検討した。その結果、アレルギー性鼻炎ならびに慢性副鼻腔炎患者においては、IFN-gammaの発現は見られたものの、Th2タイプのIL-4やIL-5については、さらにin vitroでアレルゲン刺激を行うことにより、その発現が確認できた。 3.アレルギー性鼻炎患者と慢性副鼻腔炎患者の鼻粘膜リンパ球のサイトカインプロフィールの違いについて検討し、アレルギー性鼻炎ではTh2タイイトカインがより強く発現されている傾向が見られた。Th1タイプのサイトカインについては、採取したばかりの鼻粘膜リンパ球では両疾患患者の間に有意な差はなかったが、in virtoでアレルゲン刺激を一定の時間加えたのちに、IFN-gammaの産生がむしろアレルギー性鼻炎患者で増強する傾向にあった。 4.鼻粘膜から得られる少量のリンパ球を用いて、細胞内サイトカインを蛋白レベルで測定するサイトカインフローサイトメトリーに関しては、未だアッセイシステムを確立するに至っていない。
