皮弁生着における微小循環動態とフリーラジカルの関与の解明

研究概要

元来、皮膚は大気中の酸素に直接さらされており、更に紫外線にも暴露されているため、他臓器と比較して、酸素ストレスは極めて高いことが知られている。また、活性酸素やフリーラジカルの研究が急速に進歩してきた結果、それらは生体に対し、功罪の二面性を持っており、皮弁作成時に生じる炎症、虚血.再潅流創傷治癒の各種病態において出現していると考えられるようになったが、その詳細は明らかでない部分が多い。つまり、微小循環動態において、酸素や活性酸素、フリーラジカルの間には、微妙なバランス関係の存在が示唆されており、国内外でそれらの関係を検討した報告が散見されている。我々は、微小循環モニタリングにおいて、顕微鏡下での連続微小血流循環動態の観察とその動態解析の必要性を痛感してきたので、高速ビデオカメラシステムと流体解析システムを今年度セットアップできた。これらのシステムにより、血管幅、血流速度のみならず、乱流等、瞬時に起こる血流動態のダイナミックな変化が観察可能となった。また、アンギオテンシンIIによる血管収縮時において、スーパーオキサイドが特異的に発生するを発見し、アンギオテンシンIIとフリーラジカル、NOさらには、高血圧に関して現在研究をin vivo,in vitro実験系にて進めており、その内容は現在投稿準備中である。
今後、さらにこれらの内容について、更に検討を加えていく予定である。
