菌体表層の両親媒性物質と歯周病-歯周組織のCD14分子の役割に関する研究-

研究概要

細菌細胞壁表層の両親媒性物質は、多彩な生物学的活性を発揮する。特に、グラク陰性菌外膜の内毒素性リポ多糖(LPS)やグラム陽性菌表層のリポタイコ酸(LTA)は、代表的な両親媒性物質である。両物質はマクロファージ等の細胞膜上に発現するCD14(膜CD14,mCD14)を介して細胞を活性化するといわれている。我々は昨年(平成11年)度、ヒト歯肉線維芽細胞にはmCD14を高発現する細胞と低発現の細胞が存在し、mCD14高発現歯肉線維芽細胞では、mCD14を介して腸内細菌科のLPSや合成リピドAの刺激を捉えて、インターロイキン(IL)-8を産生することを明らかにした。今年度は、1.Bacillus subtilisのLTAもmCD14を介して、歯肉線維芽細胞を活性化するのに対して、口腔レンサ球菌Streptococcus sanguisならびにstreptococcus mutansのLTAはLPSのIL-8誘導活性を阻害するアンタゴニストとして作用すること、2.歯周病関連細菌Prevotella intermediaのLPS画分は、mCD14を欠くヒト歯髄細胞をsoluble CD14依存的に活性化するが、この活性化には転写因子AP-1が係わっていることを証明し、さらに3.最近研究の進んだ、CD14に会合してLPSのシグナル伝達を担うToll-like receptor(TLR)系と、これら両親媒性物質の係わりを研究(阪大・審良静男教授らと共同)し、これまでの報告と違って、LTAはLPSと同様にTLR4によって認識されていることを明らかにし、報告者らがP.intermediaから調製した活性糖蛋白画分PGPはTLR2によって認識されることを示唆する成績を得た。
今後さらに、歯周組織において微生物を認識し応答するCD14/TLR系の役割の解明を進めて、「菌体成分による自然免疫機構の撹乱」との視点から、歯周病発症機序に迫りたいと考えている。
