エナメル質の石灰化機構と結晶基質蛋白の相互作用に関する研究

研究概要	歯や骨の硬組織における石灰化反応は、組織固有の分化した細胞群によって囲まれた微小環境下で時間的および空間的に秩序だった過程を経て進行する。本研究では、生体内で最も大きく成長するエナメル結晶の核生成から成長過程を明らかにするなかで、生体での石灰化に基本的な結晶化の駆動力と基質蛋白の役割の解明を目指している。この目的に沿って、本年度においては透析膜を応用した結晶沈殿装置を開発し、(1)Ca供給が律速となる条件下での沈殿反応と溶液組成の定常状態を測定、(2)種晶表面に吸着したアメロジェニンの分子形状を原子間力顕微鏡でin situ観察を行い、(3)アメロジェニンの溶液内分子構造の解析に向けては、FTIR全反射吸収法により生理的な溶液条件下(温度、塩濃度、pH)でのアメロジェニンの会合体形成過程を調べ、(4)結晶・基質蛋白・分解酵素(セリンプロテアーゼの代表としてトリプシンを選択)の相互作用が石灰化反応に及ぼす影響について解析した。これらの実験結果から、エナメル質成熟化に不可欠な基質蛋白の低分子化と可溶化に関与する分解酵素活性は、溶液組成と結晶成長反応自体によって制御されていることが判明した。また、ブタおよびラット形成期エナメル質における溶液相の組成(未知のイオン種、有機酸、アミノ酸、ペプチド)の同定とその定量、ブタ形成期エナメル質については、その発生段階に従って、可溶性成分(特に、有機酸)の組織内分布について検討した。さらに、マウス臼歯歯胚の発生過程と器官培養系における表現型の推移、腫瘍(エナメル上皮線維腫)でのアメロジェニンの発現と硬組織形成への関与について総括的なまとめを行った。以上の研究成果については、論文投稿に至っている。
