研究概要	本年度はペプチドリード医薬品の開発研究における合成化学的基盤技術の確立を主目的として以下の二つの研究を行った。 (1)Aza-Pavne転位反応を用いる光学活性1,2-アミノアルコール及び非天然型アミノ酸誘導体の合成研究光学活性1,2-アミノアルコール及び、非天然型アミノ酸誘導体は、ペプチドリード医薬品の開発研究において生物活性ファルマコフォアーとして重要な化合物である。セリン誘導体と各種有機金属試薬との反応により得られるアミノアルコール体を光延反応でアジリジン誘導体へと変換し、次いでアジリジンメタノール誘導体から立体選択的に2,3-エポキシアミノ誘導体へ変換するaza-Payne転位反応を鍵反応とする光学活性1,2-アミノアルコール及び各種非天然型アミノ酸の合成研究を行った。本法は特に基質遷移状態ミミック構造を有する特異的阻害剤への応用研究も併せて検討した。 (2)アミノ酸をキラルプールとする4種の光学活性な(E)-アルケンジペプチドイソスターの立体制御合成プロセスの確立アミノ酸を出発原料としてβ-aziridiny1-α,β-enoateを鍵合成中間体とする光学活性な(E)-アルケンジペプチドイソスターの立体制御合成プロセスに詳細な検討を加えた。その結果遷移金属触媒Pd(0)による異性化反応およびメタンスルホン酸によるβ-aziridiny1-α,β-enoateの位置および立体選択的開環反応と有機銅試薬によるanti-SN2反応を組み合わせることにより、(L、L)-(L、D)-(D、D)-、(D、L)-型4種の光学活性な(E)-アルケンジペプチドイソスターの立体制御合成プロセスを確立することができた。また本法を応用してボンベシンアンタゴニストやケモカインアンタゴニスト等の抗腫瘍活性および抗HIV活性を有するペプチドの非ペプチド化研究も併せて検討した。
