研究概要	独自に検討を重ねてきた有機金属試薬の特異的反応を活用して、ペプチドリード医薬品への応用研究を目的として以下の研究を行った。 (1)Aza-Payne転位反応を用いる基質遷移状態模倣型酵素阻害剤の合成 1.HIV多剤耐性克服型プロテアーゼ阻害剤の開発を目的として、まず、既知の構造情報を基にしたコンビコンビナトリアル合成によりnMオーダーの抗HIV活性を有する新規化合物を見いだした。次いで、P1-P2部位のペプチド結合を(E)-アルケンユニットに変換することにより非ペプチド性化合物に誘導した。 2.家族性アルツハイマー型痴呆症との関連が指摘されているβ-secretaseの基質遷移状態を基にペプチド性酵素阻害剤を分子設計し、一連の誘導体を合成した。 (2)Ru-触媒によるメタセシス閉環反応の検討ペプチドのグローバル配座固定を目的として、ジスルフィド結合を有する抗腫瘍活性ペプチド類(ソマトスタチン誘導体、CXCR4拮抗剤)のCysの代わりに、アリルグリシン、ホモアリルグリシン、アレニルアラニン等を導入し、Ru触媒によるオレフィン及びアレンメタセシス閉環反応に付いて精査した。 (3)(E)-アルケン型ジペプチドイソスター(EADI)の立体制御合成法の確立と応用有機亜鉛試薬、有機銅試薬の反応性を活用して、側鎖官能基を有するEADIおよび(Z)-フルオロアルケン型ジペプチドイソスターの合成法を開発し、抗腫瘍活性、抗炎症活性、抗HIV活性、血管新生抑制活性を有する低分子リードペプチドの部分的非ペプチド化に応用し、in vitroのアッセイ系でnMオーダーの生物活性を有する数種の医薬品候補化合物を見いだした。
