ペプチドリード医薬品の実践的開発戦略研究

研究概要	前年度までのの成果を踏まえて以下の研究を実施した。 1)α-位側鎖官能基を有する立体選択的(E)-アルケン型シペプチドイソスター合成法の確立すでに本研究では有機銅試薬による還元的アルキル化反応を応用したC1〜C3ユニットの官能基導入法を確立しているが、有機亜鉛-銅複合試薬による本反応の立体制御および、各種アミノ酸側鎖に対応する官能基変換反応について詳細な検討を加えた。 2)ゲノム情報集約型低分子ペプチドライブラリーの構築と非ペプチド化 Aza-Payne転位反応による非天然型アミノ酸および非水解性リン酸化アミノ酸の立体選択的合成法、非水解性ペプチド等価体の完全立体制御合成プロセス、等を組み込んだゲノム情報集約型ライブラリーの構築を目的とした研究展開を計った。具体的には、typeII'β/γ構造を有する環状ペンタペプチドを、側鎖コンフォメーションの予測が可能な基本テンプレートとして応用した。次いで、一連のペプチド性薬物(αVβ3インテグリン拮抗剤、、CXCR4-ケモカイン受容体拮抗剤,etc.)の活性pharmacophoreの空間的スクリーニングを実施した。さらに、F-(Z)-アルケン型ジペプチドイソスターおよび多置換(E)-アルケン型ジペプチドイソスターの導入による非ペプチド化および、立体配置座固定法に関して詳細の検討を加えた。 3)癌、エイズ、アルツハイマー型痴呆症等の新規化学療法剤の開発上記難治性疾患に対する独自に見いだしたペプチド性活性物質の医薬への応用を視野に入れて、当該研究の方法論を検証した。
