変異遺伝子の組織細胞での正常化に基づくA群色素性乾皮症遺伝子治療の基礎研究

研究概要	変異遺伝子の正常化には、染色体の特定部位に遺伝子を挿入することが必要である。そのためにDNA/RNAキメラ分子によるA群色素性乾皮症の変異の修正実験を試みることにした。A群色素性乾皮症の患者細胞XP20SSVの変異は第3イントロンの3'acceptor siteにおけるG->C変換が原因でフレームシフトをおこしたものであるのでこの部位を含むDNA/RNAキメラオリゴヌクレオチド(68mer)を合成し、HVJ-cationic liposomeで患者細胞に導入したが、変異は修正されなかった。次に、レトロウイルスのインテグラーゼとLTRを組み合わせた標的導入法の開発を試みた。マウス白血病ウイルスのインテグラーゼの組み換え体を作成し、これをフォスフォセルロースカラムで精製し、これをLTRを含む遺伝子とmixして複合体をつくらせ、その複合体をHVJ-リポソームで細胞内に導入したが、効率のよい遺伝子挿入はおこらなかった。最後に、魚類のトランスポゾンとトランスポゼースのない場合に比べて数十倍上昇する。しかもCationic liposomeによるlipofectionよりもHVJ-cationic liposomeによる方が数倍効果があった。そこでHVJ-anionic liposomeによりneo遺伝子をマウス肝臓に導入し、経時的にneo遺伝子の有無をPCRによって調べた。まだ解析中であるが、現在までに、トランスポゼースのない場合には、2週間後にneo遺伝子が検出できなくなったが、トランスポゼースとの共導入個体では6週間後においてもneo遺伝子が検出された。このシステムは非分裂細胞の多い組織中でも働く可能性があるので、導遺伝子のデザインを考案することによって標的部位導入が可能かもしれない。
