研究概要

本研究は、大腸菌のシャペロニンGroEL/ESの機能発現の分子メカニズムを明らかとすることを目的とする。そのため、ラクトアノレブミン(αLA)、スタフィロコッカル・ヌクレアーゼ(SNase)、グリーン蛍光タンパク質(GFP)などの物理的性質の大きく異なる球状タンパク質の中間体とシャペロニンとの相互作用をストップトフロー蛍光スペクトル法を用いて調べる。αLAについては既に昨年度までの研究でいくつかの成果を上げているが、本年はさらに、SNaseとGFPについての研究を進めた。
申請者の研究室では、SNaseのすべてのプロリン残基を他のアミノ酸に置換し、プロリン異性化の影響のないSNase(-pro)がすでに作られていた。SNase(-pro)にAla69ThrまたはAla90Serの変異を導入した二種の変異タンパク質を作成した。当初、これらの変異体は単一exponentialで巻き戻ると期待されたが、現実には2相的となった。しかし、野生型のSNaseに比べると巻き戻り速度過程が著しく単純化されており、シャぺロニンGroELの作用機構を調べる上でよいモデル標的タンパク質となることがわかった。これらの変異SNaseの巻き戻りの速度過程に及ぼすシャぺロニンGroELの影響をストップトフロー蛍光スペクトルを用いて調べた。その結果、SNaseに対するGroELのモル比が10%程度のところで、今まで知られていた速度過程よりも速い過程が観測された。タンパク質濃度依存の測定から、この過程は1分子過程であることもわかった。
GFPについては、CLONTECH社より購入したプラスミドpGFPよりGFP遺伝子を切り出し、T7プロモータを持った大腸菌プラスミドpSCREEN1-(b+)に接続し大量発現プラスミドを作成した。野生型GFPの塩酸グアニジンによるアンフォールディングを調べたところ、転移領域では反応はほとんど不可逆であった。可逆に巻き戻るGFPを作成するため、現在、いくつかの変異体を作成中である。
