研究概要	本研究の対象蛋白質の1つであるテイルリゾチームは昨年明らかになったように、351Serと352Alaの間でプロセッシングが起こる。C末端側ドメインは3量体形成に必須であり、切断後も複合体として留まる。このC末端側ドメインはテイルおよび成熟ファージ中にも存在していることがウェスタンブロッティングにより証明されていたが、テイルから直接同ドメインを単離して確認することは出来ていなかった。しかし、最近になってテイルから同タンパク質のバンドを取り出してエドマン分解により直接同C末端ドメインが存在することを証明することができた。当研究室で学位取得後米国パーデュー大学に留学した金丸は、最近gp5-gp27が複合体の結晶化に成功し、同結晶が分解能約2.5A分解能の回折点を与えることを見いだした。現在セレノメチオニン置換体を作成中である。この複合体は3分子のgp5と6分子のgp27からなるヘテロ9量体で、成功すればセレノメチオニンを用いて位相が決定できた最も大きな複合体になるとのことである。他方、小尾繊維の繊維形成に必須のgp57Aは熱変性および変性剤による変化が高度に可逆的である珍しい例である。この蛋白質の会合の性質を詳細に調べるためにUVモニターとレイリー干渉計を用いて広範囲の濃度領域で重量平均浮揚分子量を測定した。この実験は、またいくつかの温度およびいくつかの濃度のグアニジン塩酸存在下でも行った。その結果、単量体は二次構造を持たないこと、基本会合体は2量体であって六量体と平衡関係にあることが明らかとなった。高濃度領域では6量体はさらに会合する傾向にある。単量体-2量体間の平衡定数は30℃付近に極大値を持つが、2量体-6量体間の平衡定数は温度に対して単調に減少した。
