FGF,WNTシグナルによる肢形成位置情報制御

研究概要	ショウジョウバエの肢の形成において、成虫原基上での様々なモルフォンゲ・シグナルが重要な役割を果たしていると考えられている。昨年度我々は、Wingless モルフォゲンに対する新規の受容体Dfrizzled-3を単離した。様々な分子生物学的研究から、Dfrizzled-3は、Wingless の受容体ではあるが、通常の受容体Dfrizzled-2とは異なり、シグナル伝達活性は非常に弱く、またWinglessシグナルの下流でそれをコードする遺伝子が転写活性化されることを示した。さらにWingless活性の低下により触角が形成されなくなる異変を、その欠失によりrescueできることから、Winglessシグナリングを適当なレベルまでに減衰する役割をもっていると提案した。本年度は、この結果を分子遺伝的な手法で更に確証することを試みた。更に、Dfrizzled-3とは異なる肢Winglessシグナル下流遺伝子を探索し、新たに2つの受容体を見出した。一つは、受容体型チロシンキナーゼで、他は、受容体型フォスファターゼであった。いずれのリガンドも不明であるが、この発見は。肢Winglessシグナルの下流でRas pathway が働くことと、蛋白質のリン酸化脱リン酸化がWinglessシグナルの制御下にあることを強く示唆している。FGFは、脊椎動物の手足の形成に必須な因子である。ハエでは、肢形成に関わっているとの証拠はまだないが、気管形成には必須な因子であることが知られている。以前の実験でFGF受容体Breathlessの転写制御領域200bpを単離していた。そこで、この領域を詳しく調べ直し、そこにRas pathwayの下流で働く Yanリプレッサー等の結合領域があることを個体レベルでのエンハンサー・アッセイと通じて特定した。更に、FGF受容体の発現が、そのリガンドであるBranchless により正に制御されていることを明らかにした。これは、レガンドによる生の制御機構を示す大きな証拠である。
