原核・真核生物に普遍的な二成分リン酸リレー情報伝達ネットワークの分子基盤解析

研究概要	本研究計画に基づいて、分裂酵母のストレス応答とそれに関わるシグナル伝達機構に関して広範な解析を継続・展開する中で次のような多くの成果を上げてきた。 1)分裂酵母の浸透圧ストレス感受性変異株群の取得と解析を行い、多くの遺伝子を同定した。 2)分裂酵母の浸透圧ストレス応答遺伝子Gpd1(グリセロール合成遺伝子)をクローニングした。 3)Gpd1遺伝子のプロモーター構造を明らかにし、その発現がWis1-Sty1MAPキナーゼカスケードに支配されていることを示した。 4)Wis1-Sty1MAPキナーゼの解析を行い、このシグナル伝達系が多様なストレス応答に関与していることを示唆した。 5)転写因子Atf1の示すカルシュウム感受性を見出し、その機能を解析した。 6)Wis-Sty1MAPキナーゼと関連して働く転写因子群(Atf1、Atf20、Atf30)を取得・解析した。 7)分裂酵母にから新規な転写因子をコードする遺伝子(Prr1)をクローン化して、バクテリア型His-Aspリン酸リレーシグナル伝達ドメインを持つことなどの構造的特徴を明らかにした。 8)Prr1欠失変異株を作成し、Prr1がカタラーゼ遺伝子(Crr1)の発現を制御するなど酸化ストレス応答に重要であることを明らかにした。この過程でストレス応答転写因子であるPrr1欠失変異株が接合・減数分裂不能の表現型を示すことを最近になって新たに見出した。このことから、Prr1が分裂酵母における多様なストレス応答(酸化ストレスや栄養飢餓)に関わるユニークな因子であることを明らかにした。Prr1の機能やそれに関わるシグナル伝達機構を解析することで、細胞周期・減数分裂を制御するシグナル伝達機構の解析という普遍的課題に発展させることができると考えられる。
