研究概要

本研究により以下の成果が得られ、RNAヘリケースAをはじめとする転写コアクチベーター機能の分子機構、およびそれらの生物学的・医学的意義の一端を明らかにすることができた。
(1)家族性乳癌原因遺伝子産物BRCA1の転写活性化にRHAとの複合体形成が必須であることの証明(Nature Genetics 1998)。
(2)蛋白質糖化の転写因子Fosの活性化経路を報告(FASEBJ.1999)。
(3)血球系転写因子GATA1の転写活性化機構におけるCBPの重要性(PNAS 1998)。
(4)I型自己免疫性多発腺症候群(APS1)の原因遺伝子産物AIREの転写活性化へのCBPとの複合体形成の必要性(JBC 2000 in press)
(5)RHAの最小転写活性化領域の同定と同領域とATPase領域のcooperativity(投稿中)。
(6)RHAの核内輸送の分子機構(投稿中)。
(7)新潟大学辻省次教授との共同研究によりポリグルタミン病の神経変性への転写コファクターTAF135の関与を明らかにした(Nature Genetics誌 revision中)。
(8)転写統合装置レベルでの転写因子β-cateninによる細胞癌化モデルの提唱(Nature誌 revision中)。
(9)動脈硬化症病巣にて転写コアクチベータ機能の亢進状態である"hypernuclearacetylation"を検出(BBRC,1999)。
