アルツハイマー病の分子病態学的研究-βアミロイドとプレセニリンの役割

研究概要	家族性アルツハイマー病原因遺伝子産物プレセニリン蛋白の変異がアルツハイマー病脳に蓄積するアミロイドβペプチド(Aβ)の産生に及ぼす影響について研究を行った。本年度はプレセニリン2を主な対象とし、N141lアルツハイマー変異を有する分子のカルボキシ及びアミノ末端側に様々な欠損や修飾を加え、培養細胞に発現し、分泌されるAβを測定すると同時に、プレセニリン蛋白のプロセシングと安定性を検討した。正常に産生されるプレセニリン2アミノ末端断片に相当するタンパク質はN141l変異を有していてもAβ42の増加効果を示さず、カルボキシ末端数残基程度の微小な欠損を加えたのみでも、全長分子のもつAβ42増大効果は抑制された。アミノ末端側70残基以内の欠損はAβ42効果に影響を及ぼさなかった。またAβ42効果はプレセニリン2蛋白の安定化とプロセシングが起こった場合に生じていることが判明した。このことから、安定化を受けた断片型プレセニリンが活性型分子であること、プレセニリンの最C末端に安定化とAβ342効果にかかわる重要な機能ドメインが存在することが明らかとなった。さらにプレセニリン変異のアミロイド蓄積に及ぼす影響をin vivoで知るため、変異プレセニリン1と変異APPを二重に過剰発現したトランスジェニックマウスの脳組織を観察した。APP単独発現マウスに比してアミロイドの蓄積は顕著に増大したが、12ケ月齢に至るまで大脳皮質の神経細胞数に減少は見られなかった。このことはβアミロイド蓄積は神経細胞死を生じる十分条件ではないこと、アルツハイマー病脳における細胞脱落に他の因子が必要であることを示唆する。
