小脳顆粒細胞のアポトーシスに関する分子細胞生物学的研究

研究概要	小脳顆粒細胞および大脳皮質ニューロンの培養系を用いて神経栄養因子、特に脳由来神経栄養因子(BDNF)の細胞死抑制効果の解析を行った。培養大脳皮質ニューロンでは、インシュリン様成長因子-1(IGF-1)によっても、アポトーシスが顕著に抑制される。BDNFはこのアポトーシスに対し抑制効果をもつものの、その効果はIGF-1と比べ著しく低くかった。しかしながら、PI3-キナーゼの活性化およびAkt-キナーゼの活性化はほぼ同程度であった。さらにPI3-キナーゼの特異的阻害剤であるLY294002により、BDNFによる細胞死抑制効果は完全に抑えられるのに対し、IGF-1による効果は部分的にしか抑えられない。このことは、BDNFによる細胞死抑制効果はPI3-キナーゼの活性化に依存しているのに対し、IGF-1の抑制効果はPI3-キナーゼの活性化だけではなく、他の機構にも依存していることを示している。実際、PI3-K経路以外のシグナル蛋白質、Shp-2、Grb-2、N-Shc、PLC、MARK等については、両因子による活性化に大きな違いを見出している。以上のことは、アポトーシスを防御する2つの神経栄養因子間で、それらの防御機構において大きな違いが存在することを示している。今後、その違いの原因を分子レベルで明らかにすることは、神経栄養因子によるアポトーシス防御機構の解明に大きく貢献するものと考えられる。
