コンディショナルターゲッティングを用いたシナプス可塑性の分子機構解析

研究概要	本研究の目的は、コンディショナルにノックアウトする方法を開発し、シナプス可塑性成立に関わる分子が脳高次機能にどのように関与するかを個体レベルで解明することである。このために、コンディショナルターゲッティング法の開発と129系統以外の近交系マウス由来のES細胞株の樹立を進めてきた。これまでに、C57BL/6系マウス由来のES細胞株を用いて、高効率に生殖系列遺伝するキメラ作成法を実用化することに成功した(BBRC印刷中)。これまでは、キメラ作成能などから脳に奇形のある129系統由来のES細胞株の使用から脱却できないできた。学習能力が高く脳機能解析のための行動実験に実績があるC57BL/6系由来ES細胞株を用いたシステムが実用化できたことにより、膨大な時間と労力をかけて戻し交配をおこない遺伝子背景を変換して解析する必要がなくなった点は画期的である。また、プロゲステロン受容体ホルモン結合領域と遺伝子組換え酵素Creリコンビナーゼ融合タンパクであるCrePR遺伝子を用いることにより、RU486などの薬剤投与による遺伝子組換えがマウス個体で誘導出来ることを示した。さらに、小脳顆粒細胞、プルキンエ細胞、海馬CA3錐体細胞など、特定の細胞群に特異的な発現をする遺伝子プロモーターの制御下でCrePR遺伝子を発現するマウスを作成した。その成果の一部である、プルキンエ細胞特異的に組換えを誘導できるシステムについてはすでに報告した(BBRC印刷中)。また、標的とする分子として、NMDA受容体ε2サブユニット、細胞接着分子であるβカテニンなどいくつかのシナプス機能分子を選択し、それぞれの遺伝子にloxP配列を組み込んだマウスをC57BL/6系統由来ES細胞株を用いて樹立した。これらマウスの交配と薬剤投与による部位時期特異的な分子欠損マウスの作成が可能になった。
