研究概要	エストロゲンはエストロゲン受容体(ER)を介してさまざまな組織の発達、分化、機能等の制御に重要な役割を果たしている。ER遺伝子自身もエストロゲン応答配列(ERE)をプロモーター領域にもつエストロゲン応答遺伝子であり、その発現は転写及び転写後の両段階を含む過程により時期・組織特異的な制御を受ける。遺伝子の発現には、真核細胞核内のDNA、ヌクレオソーム、クロマチン、染色体といった階層構造が関与する。階層的な遺伝子制御に大きな影響を及ぼす一因にDNAの折れ曲がり構造がある。circular pemutation assayによりER遺伝子5'上流のプロモーター領域3kbに折れ曲がり構造を同定した。この領域に同定された5カ所の折れ曲がり(ERB-4からERB+1)のうち、3カ所はヌクレオソーム構造から予測される周期に合致したが、ほかの2カ所は合致しなかった。いずれも潜在的な折れ曲がり中心配列であるA2N8A2N8A2(A/A/A)を含み、短いpol(dA)・poly(dT)配列を伴っていた。折れ曲がりの一つであるERB-2の詳細なマッピングから、このA/A/A配列と、エストロゲン応答配列の半分(half ERE motif)を含むすぐ隣接する20bpの配列が、折れ曲がりに関与することが判明した。ルシフェラーゼをレポーター遺伝子として用いた転写活性実験により、この配列がエストロゲン反応性を持つことを示した。さらに、この配列に変異を起こした実験により、エストロゲン反応性はhalf ERE motifに隣接したA4 tractに関係することを明らかにした。このhalf ERE motifとmutantの一つであるGGTTAは核タンパクに対し同様の親和性を示した。これらの結果から、周期性DNA折れ曲がり構造がER発現の転写調節因子として働いていると結論した。
