野生マウス遺伝的背景の進化学的位置と生物機能モデル遺伝子発現への関与

研究概要

3種類の亜種グループから育成した8野生由来系統を対象に、1000種類のマイクロサテライトDNA,mtDNA,Y染色体上のSry等の多型を解析し、我々がこれまでに主張してきた3つの亜種グループの分離が明確であることを確認した(米川)。DDBJのデータベースから22座位の遺伝子DNAの塩基配列を野生由来8系統の間で比較した結果も上記と同じ3群の分離を示した(斉藤)。Gene-scan装置とアガロース電気泳動法の二つで95種のマイクロサテライトDNAの分析を行い17系統を比較した結果も同じ結論となった(山口、颯田、森脇)。Major satellite DNAの染色体上の分布の変異をFISHを用いて調べた結果,北東アジアのマウスを大きく5つのパターンに分けることが出来た。(松田)北海道内のマウスはmitochondria DNAから見ると道東地域にmusculus型が分布し,中央部にはcastaneus型が広く分布している。しかし、Sry遺伝子から見ると殆どmusculus型であった(鈴木)。
新しいw1型ベータヘモグロビンのイントロンの分析から、dとw1の分岐は約200万年、dとw1の組替えは20-30万年前と推定される(山口,颯田,森脇)。インドのマイソールで捕獲した野生マウスから育MYS/Mz近交系を育成したが,この系統は第1染色体上のpep3酵素活性を欠損しており劣性遺伝を示す(土屋)。カザフスタン産個体の中から新しいヘモグロビンベータ遺伝子変異Hbb-w2が見出されたが、DNA塩基の分析からd型とw1型との組替本である可能性が示唆されている。(土屋,山口、宮下)。
実験用マウスに見出された変異遺伝子Rim4を日本産野生由来MSM系統に導入するとその発現が顕著に抑制される。この修飾に関与する遺伝子が第2染色体上のIstと強く連鎖していることを明らかにした。また、短尾突然変異Ts遺伝子の本体がリボソームの構成成分L38をコードしているRol38であることを見出したが,日本産野生由来MSM系のもつRpl38遺伝子をTs系統に導入したところ,その発現が抑制された(城石)。
