IP_3受容体/Ca^<2+>放出チャネルの構造機能とIP_3/Ca^<2+>シグナル伝達に関する研究

研究概要	1. IP_3受容体チャネルは巨大な4量体複合体タンパク質(4x〜300kDa=〜1200kDa)である。この折れたたみ構造を推定するために、タイプ1IP_3受容体が豊富なマウス小脳由来のミクロゾーム画分を用いてトリプシンによる限定分解実験を行った。タイプ1IP_3受容体は5つのトリプシン耐性断片に特異的に消化されることから、大きくは5つの折れたたみドメイン構造を取ることが解った。 2. IP_3結合領域は2つのトリプシン耐性ドメインに分割されること、チャネル領域は一つのドメインにあることから、これらはコンパクトに折れたたんだ特有の高次構造が示唆される。また、アミノ酸シーケンスにより決定したトリプシン感受性部位はalternative splicing部位やカルモジュリン結合部異近傍などの調節領域にあたることから、これらの部位は比較的表面に露呈してほぐれた構造を取りやすいことが示唆された。 3. 5つのトリプシン耐性ドメインはタンパク質問相互作用によりお互いに比較的安定に会合していることが解った。トリプシン消化後のミクロゾームを用いても十分なIP_3結合活性とIP_3誘導Ca^<2+>放出活性が観察されることから、IP_3リガンド結合とチャネル開口のカップリングはこれらドメイン間の構造上の緊密な相互作用がIP_3受容体チャネルの機能に重要であることが示唆される。 4. IP3結合領域を構成する2つのトリプシン耐性ドメインの機能解析を行った。発現させたN末端側のドメインにはIP3結合活性がなく、C末端側ドメインには低親和性の活性がみられた。両者をin vitroで混合してIP3結合活性を調べたところ、高親和性の活性が現れた。このことから、これら2つのドメインによる相互作用によってIP3結合ポケットが形成されることが明らかとなった。
