研究概要	本研究は組織切片上から異常な細胞群を形態学的に観察し、その細胞内ゲノム遺伝子あるいはメッセージを直接採取しDNAシーケンスすることによって病態をより迅速に判定しうるシステムを確立することにある。本年度は組織切片からの遺伝子抽出に有効な手法の確立をめざした。その結果、1枚のパラフィン切片からも非常に効率良く遺伝子増幅の可能な手法を確立した。その際、組織の固定条件によっては目的の遺伝子の単離は不可能であることも確認された。さらに顕微鏡下にて目的の細胞を採取するため、マイクロダイセクション法を利用しウレタン誘発肺腫瘍組織からゲノム遺伝子を抽出することに成功した。本法は30μm径に集蔟する目的の細胞をレーザーによってプレパラート上から剥がし取るために開発されたもので、本研究には有効な手段であることが確認された。これらの成績から遺伝子解析をよりin situで行う端緒が築かれた。また、これらの方法を利用する疾患モデルとして、レニンノックアウトマウス、糖転移酵素トランスジェニックマウス、先天的白内障モデルマウス、精巣内減数分裂異常マウス、雄性不妊マウスなどの形態解析を行い、マイクロダイセクションの利用価値の高い組織の検索に努めた。さらに、形態学的に異常の検出できない細胞から遺伝子の微量な変化を検出する手段として、ホストゲノムサブトラクション法を試行した。本実験にはマレック病潜伏感染細胞を用い、ウイルスDNAを調整後Repr esentation Difference Analysis変法を行った。その結果、未報告のゲノム多型を検出することに成功し、ウイルス株間で起こる培養細胞の微妙な形態変化をゲノム多型から解析できる端緒を築くことができ、本方法の株間多型解析における有用性が示唆された。
