Axinによるβ-cateninの分解機構の解析と癌遺伝子診断への応用

研究概要	APC(Adenomatous polyposis coli)は、家族性大腸腺種症の原因遺伝子として単離されたが、一般の非遺伝性の大腸癌でも変異が認められることやAPCがβ-cateninと結合しβ-cateninの分解を促進する作用があることが明らかにされた。APCに変異があるとβ-cateninの分解が止まって細胞内に蓄積し転写因子Tcfが活性化されて遺伝子発現が促進されて細胞増殖が起こるということが癌化の一つのメカニズムであることが見出されている。したがって、β-cateninの分解機構を解明することは、癌の病態理解に直結すると考えられる。大腸癌や悪性黒色種の培養細胞では、β-catenin上のGSK-3βによるリン酸化部位の変異が報告されており、実験的に同じ部位を変異させるとβ-cateninの分解が阻害されることから、β-cateninの分解にはGSK-3βによるリン酸化が必須だと考えられている。私共がGSK-3β結合蛋白質として見出したAxinはAPCやGSK-3βとβ-cateninと結合し、Axin存在下ではGSK-3βによるβ-cateninのリン酸化が著明に亢進した。したがって、この4者が複合体を形成することによりβ-cateninの分解が制御されると考えられた。事実、動物細胞にAxinを発現させるとβ-cateninの分解が促進され細胞増殖が抑制された。これらの結果から、Axinは抗癌遺伝子として機能する可能性が高く、現在種々の癌組織においてAxin遺伝子の異常の検出を試みている。
