研究概要	浸潤性の癌細胞は、運動方向の細胞外基質に対して接着と分解を繰り返すことによって、浸潤性移動を達成する。この際に、接着と分解に関わる分子は浸潤先進部に配置され、細胞移動と協調して働くように制御されていると考えられる。浸潤性メラノーマ細胞をフィブロネクチンでコートしたゼラチンゲルの上におくと、接着面に対して突起を伸ばす。この時、接着面の分解はこの突起状構造の周辺から起こることが観察されている。我々はMT1-MMPがこの浸潤性突起に局在することを観察して既に報告している。今回、MT1-MMPの浸潤性突起への局在の仕組みを解析するために、MT1-MMPの細胞外ドメインをGFPに置き換えたキメラを作成して、メラノーマ細胞に発現させ、その局在を調べた。GFP/MT1はプラスチックディッシュ上では細胞全体に分散していたが、ゼラチン・フィブロネクチン上では細胞・基質接着面で垂直方向に棒状に局在することが観察された。アクチンを染色すると同様の局在を示し、MT1-MMPがアクチンと共存する可能性が示された。また、同様の実験系で浸潤突起を観察すると、突起構造はアクチン束の局在と一致し、その周辺での細胞外基質の分解像が観察された。一方で、非浸潤性のCHO細胞は浸潤突起を形成せず、よく発達したアクチン・ストレスファイバーを形成して細胞外基質に接着する。MT1-MMPをトランスフェクションにより強制発現すると、アクチン・ストレスファイバーに沿った分解像が観察された。この結果から、MT1-MMPはアクチンと相互作用があり、浸潤突起形成の際にもアクチン束形成に伴ってそこに局在すると考えられる。
