肝細胞増殖因子(HGF)の医薬への応用に向けての活性制御因子の生体内動態の解析

研究概要

肝細胞増殖因子(HGF)は、肝細胞に対する強力なマイトジェンであり、現在肝炎、肝硬変などの肝疾患に対する治療薬として開発が進められている。HGFは肝障害時にセリンプロテアーゼの1つであるHGF activator(HGFA)により活性化され肝再生因子として機能する。本研究では肝障害時におけるHGFAの活性化および不活性化について解析し以下の結果を得た。
1. 肝障害時におけるHGFAの動態について、四塩化炭素により肝障害を惹起したラットの肝臓について解析したところ、HGFAのmRNA量が肝障害に伴って増加することが明らかになった。またHGFAは通常は前駆体で血中を循環しているが、肝障害に伴ってHGFAが肝臓において活性化されることが明らかになった。さらにHGFAの活性化はトロンビンにより起こることがわかった。したがってHGFAは肝障害に伴って量が増加するとともに、血液凝固系と連動して活性化されHGFの活性制御に重要な役割を果たしていることが明らかになった。
2. HGFAの活性を阻害するインヒビターとして2種類のHGFA inhibitor(HAI)が同定されている。cDNAクローニングによりこれらのHAIは膜結合型として合成され蛋白質プロセシングにより細胞外に分泌されると推定されている。1型HAIについて抗体を作成し、イムノブロット法で動態を解析したところ、細胞膜に結合した1型HAIが同定された。また少なくとも3カ所の切断部位により切断され細胞外に分泌されてくることが明らかになった。したがってHGFAの不活性化の制御には、分泌型HAIを生成するプロセシング機構が重要であることが示唆された。
