研究概要	過剰の一酸化窒素(NO)はマクロファージなどの細胞にアポトーシスを誘導することが知られている。NOはNO合成酵素(NOS)の働きにより、アルギニンを基質として合成されるが、アルギニンはアルギナーゼの基質でもある。そのため、両者が同一の細胞に発現した場合は、基質を結合し、NO産生が抑制される可能性がある。アルギアーゼには肝型(I型)と、非肝型(II型)の二つのアイソフォームが存在する。我々はこれまでに、大腸菌リポポリサッカライド(LPS)刺激したラットの腹腔マクロファージにおいてI型アルギナーゼが、またマウスマクロファージ系RAW細胞ではII型アルギナーゼが誘導型NOS(iNOS)と共誘導されることを明らかにした。さらにLPSを腹腔内投与したラットの肺ではI型、腎臓ではII型アルギナーゼが誘導される。そこで今回RAW細胞を用いてアルギナーゼの誘導とNO産生およびアポトーシスとの関係を調べた。RAW細胞LPSとinterferon-γ(IFNγ)で刺激すると、iNOSが誘導され、大量のNOが産生され、細胞はアポトーシスを起こした。さらにデキサメサゾンとcAMPを加えたところ、iNOSに加えてII型アルギナーゼが誘導された。この時、NO産生は低下し、アポトーシスは抑制された。RAW細胞にI型あるいはII型アルギナーゼの発現プラスミドを導入して強制発現させた後LPSとIFNγで刺激すると、一定の割合(導入効率に相当する)の細胞がアポトーシスを起こさなかった。これらの結果より、RAW細胞においてiNOSとアルギナーゼとが共に発現すると、基質であるアルギニンの競合が起こり、NO産生が抑制され、アポトーシスが回避されることが明らかとなった。
