研究概要

アスピリンを含む非ステロイド性抗炎症薬は、シクロオキシゲナーゼ(COX)を阻害して炎症誘起性のプロスタグランジン(PG)の合成を抑制する。現在知られている2つのCOXアイソザイムの非ステロイド或いは、ステロイド性抗炎症薬による抑制機構と炎症との関連を明らかにし、得られた知見を胃腸障害のない新たな抗炎症薬開発に応用することを目的として研究を行い、下記の成果を得た。
1)15d-PGJ_2によるCOX-2遺伝子の発現抑制がU937細胞でみとめられるが、血管内皮細胞では抑制が認められないことと、U937細胞に比べPPARγの発現量が低い血管内皮細胞にPPARγ発現ベクターを導入すると、転写抑制が認められることを見出した。また、ヒト口腔上皮ガン細胞において抗ガン作用を示すミツバチプロポリス由来のポリフェノール化合物カフェイン酸フェネチルエステルが、ホルボールエステルによるCOX-2の誘導を抑制することを見出し、その機構を明らかにした。2)マウス骨芽細胞MC3T3-E1では、グルココルチコイドにより抑制されるCOX-2を含む何らかの蛋白が関与する、TNFαによるPGI_2受容体の誘導を見出した。また、ホップ抽出物中のフムロンが約30nMのIC_<50>でNFκBやNF-IL6の転写活性とともにCOX-2の誘導を抑制することを見出した。3)NIH3T3を使ってCOX-1とCOX-2の片方のみを発現する系を確立し、両アイソザイムのmRNAと蛋白量を調べた結果、細胞中に両アイソザイムの活性阻害物質が存在することを見出した。4)アラキドン酸を供給するsPLA_2-IIDとsPLA_2-XがCOX(特にCOX-2)と機能関連し得る分子種であることを新たに明らかにした。5)COX-1とCOX-2の活性を同レベルで抑制し、胃腸障害のない阻害剤を見出した。
