研究概要	本研究は、粘膜免疫機構から炎症性、アレルギー性消化管障害の病因病態の解明とその治療法の開発を目的に、(1)経口的に摂取された抗原物質に対する粘膜免疫機構を介した免疫反応の調節機構と、その破綻に基づく炎症性ならびにアレルギー性腸管障害の発症機序を解明し、(2)粘膜免疫反応の調節機構の一つである経口免疫寛容を利用した粘膜ワクチンによる発症の予防と治療法を提言、(3)それに必要なアレルゲンの構造決定と分離精製を行う。さらに、(4)消化管粘膜の炎症性、アレルギー性疾患の際、粘膜に浸潤した好中球とマクロファージによる粘膜障害機序の解明とそれらが産生する活性物質を不活化することを目的とした治療法提言し、実用化へのプロトコールを作製することを試みている。今年度は、粘膜免疫機構を担う腸管リンパ装置の発育を、T細胞の分化から観察し、新たにcryptfpatchの存在を発見し、また粘膜免疫機構の神経ペプチドによる制御を司る下垂体刺激ホルモン(CRT)系のurocortinの産生を腸管粘膜マクロファージに始めて証明した。その結果、こうした粘膜免疫調節系の分化成熟の不均等や発育不全が粘膜障害を惹起させるものと考えられた。更に、粘膜誘導型寛容におけるTh1及び胞Th2の関与についてIFN γ遺伝子を選択的に欠損するマウス(IFN γ^+)を用い解析を試みTh1型サイトカイント特にIFN-γが経口免疫寛容誘導・制御に重要な役割を果していることが明らかとなった。さらに、トランスジェニクマウス(OVA-TCR-Tg)を用いた経口摂取アレルゲンに対するIgE応答モデルにおいて消化管においても食物アレルギー様変化が観察され、これは、アレルギーモデルとして有用性であることが証明され今後の実験研究推進に貢献するものと思われた。
