オステオポンチンによる生体防御反応の制御機構解析とそれに基づく難病診断・治療戦略

研究概要	1.我々が樹立したオステオポンチン(OPN)遺伝子導入マウス(TGマウス)を用いて動脈硬化とOPNの関係を明らかにした。免疫系細胞に選択的に外来性OPNを発現するTGマウスを3ヶ月にわたって高脂肪食および普通食を経口摂取させ、大動脈Valsalvaにおける動脈硬化を検討した。TGおよび対照群において高脂肪食投与の有無により血中OPN値に有意差は認められなかった。また高脂肪食投与によりTG、対照群とも有意に血中コレストロール値は上昇したが、TG群に差は認められなかった。しかしながら、高脂肪食投与によりTG群では動脈硬化の面積の増大が観察され、しかも硬化壁中の活性化マクロファージに強いOPNの発現が認められた。動脈硬化とOPNの関係は、糖尿病患者の動脈壁に強くOPNが発現することからも明らかである。さらにOPNは血管平滑筋の細胞遊走とPDGFによる細胞増殖を増強することを見出した。 2.OPNと腫瘍浸潤および肺転移能との関係を明らかにした。 StageIの肺腺癌患者の予後判定因子につき検討した。OPNとVEGFの両者が陽性群は他の群とD34陽性比較し、有意にCD34陽性の血管新生が増強し、かつ生存率が有意に低下した。さらにOPNが種瘍細胞の浸潤、転移を促進するメカニズムにつき検討した。転移能が高い腺癌細胞はCD44vを発現する。細胞上のCD44vは少なくとも2種類のβ_1インテグリン分子と結合する。α_9β_1とα_?とβ_1である。CD44vと結合したβ_1インテグリンは活性化し、OPN分子中の古典的細胞接着ドメインであるRGD配列以外のアミノ酸を認識しOPNと強く結合する。 3.OPNを認識する特異的抗体およびELISAを作製した。 OPNは分子内にトロンビンで特異的に切断される部位をもち、炎症部位では断片化する。断片化したOPNを測定できる抗体、ELISAはこれまで存在しなかったが、我々は初めてELISAを樹立した。
