原虫感染症の防御の本体-胸腺外分化T細胞の活性化の意味

研究概要	原虫感染症はその細胞寄生ゆえに、系統発生学的に進化レベルの高い通常の胸腺由来T細胞やB細胞の産出する抗体が攻撃できない状態で寄生に成功している。では、まったく生体が無防備かというと一定の抵抗性もある。このような抵抗性が胸腺外分化T細胞によって行われていると思われるようなデータが筆者らの研究から生まれている。ヒトやマウスのマラリア感染症(平成9年度寄生虫学会シンポジウムで発表)、ヒトトリコモナス症、カリニ肺炎などで胸腺外分化T細胞の著しい活性化が認められたからである。これらの知見を元に本研究では、自己応答性を持つ胸腺外分化T細胞が原虫感染一般でどのような生体防御系として働いているかを明らかにしてゆく。主に、マウスにPlasmodium(P.)yoeliiを感染させることで、原虫感染のエフェクター細胞の主体を明らかにできた。感染(blood stage)後3〜7日目に肝と脾を中心にして激しい炎症がありCD3^<int>細胞(マウス胸腺外分化T細胞)の著しい増加が認められた。この際、マラリア原虫感染赤血球はMHC class I(+)でありこれがCD3^<int>細胞の標的と考えられた。CD3^<int>細胞にはNK1.1^+(NKT細胞)とNK1.1^-のサブセットがあるがNK1.1^-サブセットがより関与していると思われた。
