研究概要	平成10年度は、これまでのNF-κB活性化機構の研究をさらに進め、新たにNF-κBの生理的な阻害分子であるIκBの分解にCaシグナルのエフェクター・プロテアーゼであるカルパインが関わることを見出した。また、NF-κBの働きと調節機構を明らかにするために、NF-κBp65 subunit(転写活性化ドメインを欠失させたもの)をbaitとし、酵母two-hybrid screening法を用いてp65と結合する蛋白分子の遺伝子クローニングを実施し、約2x10^6個のクローンの中から複数の遺伝子を新たに分離・同定した。このうちのひとつは癌抑制遺伝子産物p53と結合する性質から当初同定され、その後Bcl-2にも結合することの明らかになった53BP2であった。また、別の遺伝子は、NF-κBの転写活性化作用に対する抑制効果を持つことが明らかになったためRelA-associated inhibitor(RAI)と命名した(特許申請中)。RAIのco-repressorとしての作用機序については引き続き研究中である。培養細胞での疾患モデルを用いた研究では、すでに報告したようにNF-κBがHIVプロウイルスの賦活化に主要宿主転写因子として作用しているために、新たな化合物の有効性をHIV潜伏感染細胞で実証した。また、慢性関節リウマチ(RA)においても、RA治療薬として用いられている金製剤がNF-κBを阻害しRA滑膜細胞からのサイトカイン産生を抑制することを示した。興味深いことに、金製剤はHIV潜伏感染細胞からのウイルス増殖も抑制した。このような結果は、従来はカテゴリーの異なると考えられていた疾患が共通の転写因子によって横断的に理解され、しかも一方の疾患で有効な治療法がこの因子を標的とするならば、分子病態を共有する他の疾患に対しても有効な場合があることの例といえる。なお、NF-κBの持つ抗アポトーシス活性は、細胞のがん化(ならびに放射線・化学療法に対する耐性獲得)およびRAにおける滑膜細胞増殖にも関わっている可能性が示唆されるので、この点についてさらに実験を進めているところである。
