アレルギーの病因遺伝子群の分子遺伝学的、構造生物学的解明と治療・予防への応用

研究概要	本研究の目的は分子遺伝学的手法及び構造生物学的手法を駆使し、アレルギー素因を規定しているアレルギーの多遺伝子を世界に先駆けて系統的・総合的に解明し、さらにその遺伝子がコードしているタンパクの立体構造を解明し、治療予防に応用することである。平成12年度は平成11年度に引き続き、アレルギーの病因遺伝子群を以下の方法で系統的、多角的に検索し、世界的な知見を見出した。 IgE遺伝子とその発現異常の解明。アレルギーの最も基盤となり、かつ異常なタンパクであるIgE抗体の産生過剰をB細胞内部と外部(T細胞など)の両面から引き続き系統的に分子遺伝的に解析した。このうちB細胞外部からの検討で世界的な知見が得られた。 Th1細胞からのIFN-γの産生低下、IFN-γ産生を誘導するIL-12に着目し、その異常を遺伝子学的に解析したところ、IgE高値の患者ではIFN-γ産生が低下しており、それはmRNAのレベルで起こっており、さらにその上位にあたるIL-12の産生やIL-12への反応低下が明らかになった。この原因として、IL-12レセプターβ2鎖遺伝子の異常を世界に先駆け明らかにした。さらにin vitroでIgE産生系を作成し、検討した。結果IFN-γ低下によりIgE産生の抑制が効かないことが示された。これはアレルギーの病遺伝子の重要な1つがIgE産生抑制系の遺伝子異常であることを示した初めての成果である。さらに、IFN-γ産生を誘導するIL-18に着目し、まず(1)IL-18を大量に精製し、IL-18の立体構造を明らかにした。次いで(2)抑制系であるIL-18シグナリングにつき検討したところ、IL-18レセプターα鎖遺伝子異常によりIgE産生異常を来すことを世界で初めて明らかにした。上記の成果をもとに、アトピーを遺伝子学的に分類し、オーダーメイド治療・予防への応用を進めている。
