遺伝性皮膚角化異常症に関する研究

研究概要	皮膚の角化異常性疾患は遺伝性角化症と非遺伝性角化症に大別される。遺伝性角化症に属する疾患としては、遺伝性魚鱗癖癬、ダリエー病、掌蹠角化症、汗孔角化症など多数の疾患があるが、この中で現在遺伝子レベルでの異常が解明されているのは、水庖型先天性魚鱗癬様紅皮症、伴性遺伝性尋常性魚鱗癬、葉状魚鱗癬、一部の掌蹠角化症、シェグレンラルソン症候群、先天性爪甲肥厚症などごく一部である。他の遺伝性角化異常症においては,遺伝子レベルでの異常は全く解っていない。近年トランスグルタミナーゼ1の遺伝子異常(即ちポイントミューテーション)により、葉状魚鱗癬が生じることが明らかにされた。トランスグルタミナーゼ3はトランスグルタミナーゼ1と同様CEの形成において重要な働きをするので、トランスグルタミナーゼ3の遺伝子異常がある種の角化異常症の原因遺伝子である可能性は非常に高いと考えられる。我々の研究月的はトランスグルタミナーゼ3遺伝子のノックアウトマウスを作成し,角化異常症の動物モデルを作製することを目的とする。現在遺伝性皮膚角化異常症において動物モデルが作成され,同時に遺伝子レベルでの異常も解明されているのは、水庖型先天性魚鱗癬様紅皮症と葉状魚鱗癬のみである。この点において発生工学の手法を応用した角化異常症の動物モデルの作成は非常に独創的なものといえる。更にこの動物モデルの作成後、その疾患を持つ患者よりDNAを単離しそのポイントミューテーション等を発見することは診断、治療のみならず将来の臨床応用に多大な貢献をするものである。現在トランスグルタミナーゼ3遺伝子のターゲッテイングベクターを作製しようとしている所である。
