新しいゴナドトロピン誘導性転写因子GIOTの遺伝子診断への応用

研究概要

私共が発見した新しいゴナドトロピン誘導性転写因子GIOTを遺伝子診断に役立てるため、その卵巣での機能を明らかにすると共に、そのヒトホモローグをクローニングし解析を行った。GIOTを組み込んだ発現ベクターを卵巣顆粒膜細胞初代培養にトランスフェクトし、顆粒膜細胞内でGIOTを発現させ、ステロイド合成関連遺伝子などのプロモーター活性に対する影響を測定した。その結果、GIOTは遺伝子発現を抑制する活性を有することを明らかにした。同様に、アンドロゲン産生腫瘍細胞株MA-10を用い、GIOTを組み込んだ発現ベクターをトランスフェクトし、アンドロゲン産生腫瘍株MA-10でGIOTを発現させ、ステロイド合成関連遺伝子などのプロモーター活性に対する影響を測定した。その結果、同様にGIOTはMA10細胞での遺伝子発現に抑制的に作用する事を明らかにした。またGIOTのヒトホモローグをクロ-ニングし、そのcDNAクローンの全長の塩基配列を決定し、構造解析を行った。ヒトGIOTには、互いに類似した4つの遺伝子が存在することが明らかとなり、ヒトGIOT-1からGIOT-4と命名した。ヒトGIOTではKRABメインを持つものと持たないものとがありそれぞれの生体内での役割の違いが示唆された。現在それぞれのヒトGIOTの発現パターンや転写活性の検討を行っている。しかしながら、ヒトGIOTはラットGIOTとの相同性がかなり低いため、卵巣での役割については現在不明である。ヒトGIOTのクローニングにより、遺伝子診断への応用が可能となった。
